چکیده

The complement fragment C5a and its des-Arg74 derivative, C5a des Arg, are potent mediators of leucocyte chemotaxis and are thought to play an important role in the accumulation of leucocytes at sites of inflammation. Although specific receptors for C5a have been demonstrated on murine macrophages (Chenoweth et al., 1982) and human monocytes (Chenoweth & Goodman, 1983; Marder et al., 1985), most work to date has concentrated on the human neutrophil CSa receptor due to the relative abundance and ease of purification of this cell type. Using a monocyte-like cell line, U-937, as a model of the monocyte response to C5a des Arg, we show here the level ofCSa des Arg receptor expression appears to be regulated by the extracellular concentration of C5a des Arg itself. This may provide a mechanism by which the cell is able to maintain a chemotactic response over a wide range of attractant concentrations.

Evaluating the Effects of Cytomegalovirus Glycoprotein B on the Maturation and Function of Monocyte-derived dendritic cells

Background & Objectives: Interaction of cytomegalovirus glycoprotein B with toll-like receptors of dendritic cells leads to early signaling and innate immune responses. The aim of this study is to evaluate the effects of cytomegalovirus glycoprotein B on the maturation and function of monocyte-derived dendritic cells in treated groups in comparison with control groups. Materials & Methods: Bloo...

THE EFFECT OF HYPERTHERMIA ON THE DIFFERENTIATION OF LEUKEMIC CELL LINES

Treatment of human promonocytic leukemic cell line U937 with mild hyperthermia in the temperature range of 40-43°C resulted in differentiation of these cells into monocyte/macrophage-like cells in a heat dose and time dependent manner. This process was accompanied by marked morphological, functional and proliferational changes. U937 cells which normally grow in supension in the logarithmic...

Hydrophobic adsorption chromatography of colony-stimulating activities and erythroid-enhancing activity from the human monocyte-like cell line, GCT.

The human cell line, GCT, secretes hemopoietins into serum-free culture medium. The conditioned medium contains activities that stimulate neutrophil-monocyte, macrophage, eosinophil, and erythroid colony growth in human marrow cultures. We have used hydrophobic adsorption chromatography to separate a neutrophil-monocyte colony-stimulating factor (CSF) from the other colony-stimulating activitie...

Tumor-associated leukemia inhibitory factor and IL-6 skew monocyte differentiation into tumor-associated macrophage-like cells.

Tumor-associated macrophages (TAMs), the most abundant immunosuppressive cells in the tumor microenvironment, originate from blood monocytes and exhibit an IL-10(high)IL-12(low) M2 profile. The factors involved in TAM generation remain unidentified. We identify here leukemia inhibitory factor (LIF) and IL-6 as tumor microenvironmental factors that can promote TAM generation. Ovarian cancer asci...

Wharton’s Jelly-derived Mesenchymal Stem Cells can Differentiate into Hepatocyte-like Cells by HepG2 Cell Line Extract

Background: Wharton’s jelly is an unlimited source of stem cells that can be used in cell therapy and tissue engineering without any ethical concern. It has been revealed the cell-free extract could be effective to induce cell differentiation. The objective of this study was to induce Wharton’s jelly-derived mesenchymal stem cells (MSCs) into hepatocyte-like cells by premeabilization of the cel...
